#11 謎多き（？）タキサン系抗がん剤に迫る！＋病院薬剤師のチョイガチ細胞障害性抗がん剤ばなし タキサン編

タキサン系抗がん剤とは？
特徴的なことは？

現在、パクリタキセル、ドセタキセル、カバジタキセル、アルブミン懸濁型パクリタキセルの4つが使用されています。
おもしろポイントとしては…

  1. 植物由来！イチイから見つかった抗がん剤
    1969年、アメリカ国立がん研究所がスクリーニングプログラムによって、パクリタキセルを発見しました。
    ただ、収率が悪いこと・イチイの樹皮が必要であること（＝イチイを伐採する必要）が問題となりました。
    いまでは、イチイの葉・小枝から高収率でとれる「デアセルバッカチンIII」を使用して、半合成しています。
    → 葉・小枝から取れる！サスティナブルですね～

  2. パクリタキセル全合成も成功しているが、今も研究分野
    複雑な3次元炭素骨格、多数の酸素官能基が密集しており、合成が困難。現在でも全合成は挑戦的課題のようです。

  3. 分布容積が大きい！
    体中に広がりやすいタイプの抗がん剤！
    （脱毛などの副作用とも関連していると思う…）

  4. 脂溶性
    水に溶けにくい→アルコール and/or 有機溶媒を使用する必要がある
    → アレルギー反応、アナフィラキシーショックの原因となる。
    → 前投薬でアレルギー対策が必要。
