抗体依存性感染拡大（ADE）は、ワクチン戦略における安全性の懸念である。最近の発表では、Liら（Cell 184 :1-17, 2021）が、SARS-CoV-2スパイクタンパク質のN末端ドメイン（NTD）に向けられた感染増強抗体は、in vitroではウイルス感染を促進するが、in vivoでは促進しないことを報告しています。しかし、この研究はオリジナルのWuhan/D614G株を用いて行われたものである。

現在、Covid-19パンデミックではDelta変異体が主流となっているため、これらの変異体のNTDと促進抗体の相互作用を解析した。

分子モデリングの手法を用いて、エンハンシング抗体は、Wuhan/D614GのNTDよりもDeltaバリアントに対して高い親和性を持つことを示した。また、エンハンシング抗体は、NTDを脂質ラフトマイクロドメインに固定することで、スパイク三量体の宿主細胞膜への結合を強化することを示した。

この安定化メカニズムは、受容体結合ドメインの脱マスキングを引き起こす構造変化を促進すると考えられる。NTDは中和抗体の標的にもなることから、今回のデータは、ワクチン接種者における中和抗体と促進抗体のバランスは、オリジナルのWuhan/D614G株では中和に有利であることを示唆している。

しかし、Delta変異体の場合、中和抗体はスパイクタンパクに対する親和性が低下しているのに対し、促進抗体は顕著に親和性が上昇しています。したがって、オリジナルのWuhan株スパイク配列に基づくワクチン（mRNAまたはウイルスベクター）を接種している人にとっては、ADEが懸念されます。

It is explained very scientifically and very exact – based on data and scientific proof.

People that have taken the COVID vaccine are now subjected to ADE (Antibody-Dependent Enhancement) which is highly likely to deal grave damages to their organisms, most eventually leading to death. The absolute worst-case scenario – of the disease, as well as the vaccine. But have you ever thought that the vaccine is the plan B – to get rid of us, in case plan A – the COVI pandemia didn’t kill us already?!

On August 9, The Journal of Infection published a peer-reviewed study titled:

Antibody-dependent enhancement (ADE) of infection is a safety concern for vaccine strategies. In a recent publication, Li et al. (Cell 184 :1-17, 2021) have reported that infection-enhancing antibodies directed against the N-terminal domain (NTD) of the SARS-CoV-2 spike protein facilitate virus infection in vitro, but not in vivo. However, this study was performed with the original Wuhan/D614G strain.

Since the Covid-19 pandemic is now dominated by Delta variants, we analyzed the interaction of facilitating antibodies with the NTD of these variants.

Using molecular modeling approaches, we show that enhancing antibodies have a higher affinity for Delta variants than for Wuhan/D614G NTDs. We show that enhancing antibodies reinforce the binding of the spike trimer to the host cell membrane by clamping the NTD to lipid raft microdomains.

This stabilizing mechanism may facilitate the conformational change that induces the demasking of the receptor-binding domain. As the NTD is also targeted by neutralizing antibodies, our data suggest that the balance between neutralizing and facilitating antibodies in vaccinated individuals is in favor of neutralization for the original Wuhan/D614G strain.

However, in the case of the Delta variant, neutralizing antibodies have a decreased affinity for the spike protein, whereas facilitating antibodies display a strikingly increased affinity. Thus, ADE may be a concern for people receiving vaccines based on the original Wuhan strain spike sequence (either mRNA or viral vectors).

Antibody-Dependent Enhancement (ADE) is every virologist’s worst nightmare. It means the vaccine does the opposite of what was intended.

“In antibody-dependent enhancement, sub-optimal antibodies bind to both viruses and gamma receptors expressed on immune cells, then promoting infection of these cells.” In other words, your immune cells themselves BECOME infected and carry the virus.

Vaccinated antibodies will be a trojan horse that gives the virus entry into cells. ALL CELLS. The whole human body.

We will see massive sickness and death within the vaccinated. Boosters will likely make the situation even worse.”

Honestly, I’ve become a semi-medical person, reading so many scientific articles lately. At the first time, I used a medical vocabulary book to understand things better, watched some videos on the procedures used in medicine… But now, I am better at reading and understanding this crucial information – because I’m fighting to survive!

We have the worst-case scenario – thanks to the mRNA approach and the COVID vaccine. Thank you governments for caring about the future f the human race!
